High Mannose N-Glycans

Cell-surface oligosaccharides covalently attached to proteins and lipids are key binding epitopes in many important biological processes, including bacterial infection, cell development and the immune response. Understanding these processes at the molecular level requires access to diverse oligosaccharides of known structure and high purity to support investigations of enzyme substrate binding and specificity, to screen for carbohydrate binding proteins, and to develop assays for enzymic activity.

Omicron has focused on the synthesis of oligosaccharides derived from the parent high-mannose oligosaccharide, Man9GlcNAc2Glc3, that is transferred en-bloc to nascent polypeptides during translation in vivo. In 2014–2015, we developed a "bottom-up" approach to their chemical synthesis, leading to the initial preparation of more than 30 oligosaccharides (trisaccharide to hexasaccharides) and demonstrating the general applicability of the approach. Since that time, Omicron has prepared more than 100 high-mannose N-glycans. These products range from mannobioses (Man2) to the mannose nonasaccharide (Man9). Mannose oligosaccharides with one or two N-acetyl-glucosamine residues are also available, as well as mannose oligosaccharides containing mannose 6-phosphate residues. These products can be labeled with stable isotopes, including 2H, 13C and 15N. We can also prepare oligosaccharide sub-fragments of complex-type, sialic acid-containing N-glycans, and some of these products are listed elsewhere on this website and in our catalog.

In the following list of oligosaccharide products, all monosaccharide residues are pyranose rings having the D-configuration. Please contact us if you have questions about these products or require a product not listed here.
