Quality Assurance Assay: HPLC

Products are analyzed routinely for chemical purity by high pressure liquid chromatography (HPLC) on two instruments.

Carbohydrate Analysis

Chromatogram 1 illustrates typical resolution of glucose, mannose, and fructose. Each hexose is ~5 µg/µL. A 10 µL aqueous sample was injected on a 300 X 7.8mm Phenomenex column (Rezex 8µ 8%Ca. Monos) with a Rainin Dynamax RI-1 refractive index detector. Solvent (water) flow was 0.6 mL/min. The column temperature was regulated at 85 degrees C.

Nucleoside and Nucleotide Analysis

Chromatogram 2 is an analysis of [1'-13C]adenosine. Nucleosides and nucleotides are assayed by HPLC using a binary gradient. Sample detection is provided by a diode array UV-VIS detector.
